Non-linear mixed effects modeling and simulation (NLME M&S) is evaluated to be used for standardization with longitudinal data in presence of confounders. Standardization is a well-known method in causal inference to correct for confounding by analyzing and combining results from subgroups of patients. We show that non-linear mixed effects modeling is a particular implementation of standardization that conditions on individual parameters described by the random effects of the mixed effects model. Our motivation is that in pharmacometrics NLME M&S is routinely used to analyze clinical trials and to predict and compare potential outcomes of the same patient population under different treatment regimens. Such a comparison is a causal question sometimes referred to as causal prediction. Nonetheless, NLME M&S is rarely positioned as a method for causal prediction.
As an example, a simulated clinical trial is used that assumes treatment confounder feedback in which early outcomes can cause deviations from the planned treatment schedule. Being interested in the outcome for the hypothetical situation that patients adhere to the planned treatment schedule, we put assumptions in a causal diagram. From the causal diagram, conditional independence assumptions are derived either using latent conditional exchangeability, conditioning on the individual parameters, or using sequential conditional exchangeability, conditioning on earlier outcomes. Both conditional independencies can be used to estimate the estimand of interest, e.g., with standardization, and they give unbiased estimates.

When constructing models to summarize clinical data to be used for simulations, it is good practice to evaluate the models for their capacity to reproduce the data. This can be done by means of Visual Predictive Checks (VPC), which consist of (1) several reproductions of the original study by simulation from the model under evaluation, (2) calculating estimates of interest for each simulated study and (3) comparing the distribution of those estimates with the estimate from the original study. This procedure is a generic method that is straightforward to apply, in general. Here we consider the application of the method to time to event data and consider the special case when a time-varying covariate is not known or cannot be approximated after event time. In this case, simulations cannot be conducted beyond the end of the follow-up time (event or censoring time) in the original study. Thus, the simulations must be censored at the end of the follow-up time. Since this censoring is not random, the standard KM estimates from the simulated studies and the resulting VPC will be biased. We propose to use inverse probability of censoring weighting (IPoC) method to correct the KM estimator for the simulated studies and obtain unbiased VPCs. For analyzing the Cantos study, the IPoC weighting as described here proved valuable and enabled the generation of VPCs to qualify PKPD models for simulations. Here, we use a generated data set, which allows illustration of the different situations and evaluation against the known truth.

Emotion plays an essential role in human-to-human communication, enabling us to convey feelings such as happiness, frustration, and sincerity. While modern speech technologies rely heavily on speech recognition and natural language understanding for speech content understanding, the investigation of vocal expression is increasingly gaining attention. Key considerations for building robust emotion models include characterizing and improving the extent to which a model, given its training data distribution, is able to generalize to unseen data conditions. This work investigated a long-shot-term memory (LSTM) network and a time convolution - LSTM (TC-LSTM) to detect primitive emotion attributes such as valence, arousal, and dominance, from speech. It was observed that training with multiple datasets and using robust features improved the concordance correlation coefficient (CCC) for valence, by 30\% with respect to the baseline system. Additionally, this work investigated how emotion primitives can be used to detect categorical emotions such as happiness, disgust, contempt, anger, and surprise from neutral speech, and results indicated that arousal, followed by dominance was a better detector of such emotions.

We propose a frequentist testing procedure that maintains a defined coverage and is optimal in the sense that it gives maximal power to detect deviations from a null hypothesis when the alternative to the null hypothesis is sampled from a pre-specified distribution (the prior distribution). Selecting a prior distribution allows to tune the decision rule. This leads to an increased power, if the true data generating distribution happens to be compatible with the prior. It comes at the cost of losing power, if the data generating distribution or the observed data are incompatible with the prior. We illustrate the proposed approach for a binomial experiment, which is sufficiently simple such that the decision sets can be illustrated in figures, which should facilitate an intuitive understanding. The potential beyond the simple example will be discussed: the approach is generic in that the test is defined based on the likelihood function and the prior only. It is comparatively simple to implement and efficient to execute, since it does not rely on Minimax optimization. Conceptually it is interesting to note that for constructing the testing procedure the Bayesian posterior probability distribution is used.
