Mesoscale molecular assemblies on the cell surface, such as cilia and filopodia, integrate information, control transport and amplify signals. Synthetic devices mimicking these structures could sensitively monitor these cellular functions and direct new ones. The challenges in creating such devices, however are that they must be integrated with cells in a precise kinetically controlled process and a device's structure and its precisely structured cell interface must then be maintained during active cellular function. Here we report the ability to integrate synthetic micro-scale filaments, DNA nanotubes, into a cell's architecture by anchoring them by their ends to specific receptors on the surfaces of mammalian cells. These filaments can act as shear stress meters: how anchored nanotubes bend at the cell surface quantitatively indicates the magnitude of shear stresses between 0-2 dyn per cm2, a regime important for cell signaling. Nanotubes can also grow while anchored to cells, thus acting as dynamic components of cells. This approach to cell surface engineering, in which synthetic biomolecular assemblies are organized within existing cellular architecture, could make it possible to build new types of sensors, machines and scaffolds that can interface with, control and measure properties of cells.

This study explores the validity of chain effects of clean water, which are known as the "Mills-Reincke phenomenon," in early twentieth-century Japan. Recent studies have reported that water purifications systems are responsible for huge contributions to human capital. Although some studies have investigated the instantaneous effects of water-supply systems in pre-war Japan, little is known about the chain effects of these systems. By analyzing city-level cause-specific mortality data from 1922-1940, we find that a decline in typhoid deaths by one per 1,000 people decreased the risk of death due to non-waterborne diseases such as tuberculosis and pneumonia by 0.742-2.942 per 1,000 people. Our finding suggests that the observed Mills-Reincke phenomenon could have resulted in the relatively rapid decline in the mortality rate in early twentieth-century Japan.

Migrating cells possess intracellular gradients of Rho GTPases, but it is unknown whether these shallow gradients themselves can induce motility. Here we describe a new method to present cells with induced linear gradients of active, endogenous Rac without receptor activation. Gradients as low as 15% were sufficient to not only trigger cell migration up the synthetic gradient, but also to induce both cell polarization and repolarization. Response kinetics were inversely proportional to Rac gradient values, in agreement with a new mathematical model, suggesting a role for natural input gradient amplification upstream of Rac. Increases in Rac levels beyond a well-defined threshold dramatically augmented polarization and decreased sensitivity to the gradient value. The threshold was governed by initial cell polarity and PI3K activity, supporting a role for both in defining responsiveness to natural or synthetic Rac activation. Our methodology suggests a general way to investigate processes regulated by intracellular signaling gradients.

Depression has been associated with impaired neural processing of reward and punishment. However, to date, little is known regarding the relationship between depression and intertemporal choice for gain and loss. We compared impulsivity and inconsistency in intertemporal choice for monetary gain and loss (quantified with parameters in the q-exponential discount function based on Tsallis' statistics) between depressive patients and healthy control subjects. This examination is potentially important for advances in neuroeconomics of intertemporal choice, because depression is associated with reduced serotonergic activities in the brain. We observed that depressive patients were more impulsive and time-inconsistent in intertemporal choice action for gain and loss, in comparison to healthy controls. The usefulness of the q-exponential discount function for assessing the impaired decision-making by depressive patients was demonstrated. Furthermore, biophysical mechanisms underlying the altered intertemporal choice by depressive patients are discussed in relation to impaired serotonergic neural systems.
