Objectives: Leveraging artificial intelligence (AI) in conjunction with electronic health records (EHRs) holds transformative potential to improve healthcare. Yet, addressing bias in AI, which risks worsening healthcare disparities, cannot be overlooked. This study reviews methods to detect and mitigate diverse forms of bias in AI models developed using EHR data. Methods: We conducted a systematic review following the Preferred Reporting Items for Systematic Reviews and Meta-analyses (PRISMA) guidelines, analyzing articles from PubMed, Web of Science, and IEEE published between January 1, 2010, and Dec 17, 2023. The review identified key biases, outlined strategies for detecting and mitigating bias throughout the AI model development process, and analyzed metrics for bias assessment. Results: Of the 450 articles retrieved, 20 met our criteria, revealing six major bias types: algorithmic, confounding, implicit, measurement, selection, and temporal. The AI models were primarily developed for predictive tasks in healthcare settings. Four studies concentrated on the detection of implicit and algorithmic biases employing fairness metrics like statistical parity, equal opportunity, and predictive equity. Sixty proposed various strategies for mitigating biases, especially targeting implicit and selection biases. These strategies, evaluated through both performance (e.g., accuracy, AUROC) and fairness metrics, predominantly involved data collection and preprocessing techniques like resampling, reweighting, and transformation. Discussion: This review highlights the varied and evolving nature of strategies to address bias in EHR-based AI models, emphasizing the urgent needs for the establishment of standardized, generalizable, and interpretable methodologies to foster the creation of ethical AI systems that promote fairness and equity in healthcare.

This helps formulate hypotheses regarding molecular mechanisms that could potentially be targeted by future medicines. The CausalBench Challenge was an initiative to invite the machine learning community to advance the state of the art in constructing gene-gene interaction networks. These networks, derived from large-scale, real-world datasets of single cells under various perturbations, are crucial for understanding the causal mechanisms underlying disease biology. Using the framework provided by the CausalBench benchmark, participants were tasked with enhancing the capacity of the state of the art methods to leverage large-scale genetic perturbation data. This report provides an analysis and summary of the methods submitted during the challenge to give a partial image of the state of the art at the time of the challenge. The winning solutions significantly improved performance compared to previous baselines, establishing a new state of the art for this critical task in biology and medicine.

Label-free detecting multiple analytes in a high-throughput fashion has been one of the long-sought goals in biosensing applications. Yet, for all-optical approaches, interfacing state-of-the-art label-free techniques with microfluidics tools that can process small volumes of sample with high throughput, and with surface chemistry that grants analyte specificity, poses a critical challenge to date. Here, we introduce an optofluidic platform that brings together state-of-the-art digital holography with PDMS microfluidics by using supported lipid bilayers as a surface chemistry building block to integrate both technologies. Specifically, this platform fingerprints heterogeneous biological nanoparticle populations via a multiplexed label-free immunoaffinity assay with single particle sensitivity. Herein, we first thoroughly characterise the robustness and performance of the platform, and then apply it to profile four distinct ovarian cell-derived extracellular vesicle populations over a panel of surface protein biomarkers, thus develo** a unique biomarker fingerprint for each cell line. We foresee that our approach will find many applications where routine and multiplexed characterisation of biological nanoparticles is required.

The visual system evolved to process natural scenes, yet most of our understanding of the topology and function of visual cortex derives from studies using artificial stimuli. To gain deeper insights into visual processing of natural scenes, we utilized widefield calcium-imaging of primate V4 in response to many natural images, generating a large dataset of columnar-scale responses. We used this dataset to build a digital twin of V4 via deep learning, generating a detailed topographical map of natural image preferences at each cortical position. The map revealed clustered functional domains for specific classes of natural image features. These ranged from surface-related attributes like color and texture to shape-related features such as edges, curvature, and facial features. We validated the model-predicted domains with additional widefield calcium-imaging and single-cell resolution two-photon imaging. Our study illuminates the detailed topological organization and neural codes in V4 that represent natural scenes.
