We investigate the mechanical properties of bilayers of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine in the gel phase by using peak force tap** with quantitative nanomechanical map**. We study both dry and aqueous bilayers and liposomes supported on the oxidized silicon surface. We report the observation of a marked substrate effect on the measured Young modulus of supported bilayer stacks which decreases as the height of the stack increases. In contrast a clear substrate effect is not observed for the top bilayer of supported aqueous liposomes, which is however affected by the surface curvature of the sample. Adhesion forces present quantitative differences between dry and aqueous samples, with the former being dominated by capillary effects and the latter by non-contact interactions between tip and substrate. The mechanical properties of stacked bilayers reveal a threshold between two different regimes, for the lower portion and the upper portion of the stack, that shows a change in the plasticity of the system. The threshold is affected by applied setpoint and aging of the sample. We propose that it arises from the presence of thin layers of hydration water between the headgroups of contacting phospholipid bilayers, which appears to have a major effect on the response of the bilayer to an external mechanical stress.

This document reports an Open 2D Electrical Impedance Tomography (EIT) data set. The EIT measurements were collected from a circular body (a flat tank filled with saline) with various choices of conductive and resistive inclusions. Data are available at http://fips.fi/ EIT_dataset.php and can be freely used for scientific purposes with appropriate references to them, and to this document at https://arxiv.longhoe.net. The data set consists of (1) current patterns and voltage measurements of a circular tank containing different targets, (2) photos of the tank and targets and (3) a MATLAB-code for reading the data. MICADO will be the first-light wide-field imager for the European Extremely Large Telescope (E-ELT) and will provide difiraction limited imaging (7mas at 1.2mm) over a ~53 arcsecond field of view. In order to support various consortium activities we have developed a first version of SimCADO: an instrument simulator for MICADO. SimCADO uses the results of the detailed simulation efforts conducted for each of the separate consortium-internal work packages in order to generate a model of the optical path from source to detector readout. SimCADO is thus a tool to provide scientific context to both the science and instrument development teams who are ultimately responsible for the final design and future capabilities of the MICADO instrument. Here we present an overview of the inner workings of SimCADO and outline our plan for its further development.

On the Reproducibility of TCGA Ovarian Cancer MicroRNA Profiles

Dysregulated microRNA (miRNA) expression is a well-established feature of human cancer. However, the role of specific miRNAs in determining cancer outcomes remains unclear. Using Level 3 expression data from the Cancer Genome Atlas (TCGA), we identified 61 miRNAs that are associated with overall survival in 469 ovarian cancers profiled by microarray (p<0.01). We also identified 12 miRNAs that are associated with survival when miRNAs were profiled in the same specimens using Next Generation Sequencing (miRNA-Seq) (p<0.01). Surprisingly, only 1 miRNA transcript is associated with ovarian cancer survival in both datasets. Our analyses indicate that this discrepancy is due to the fact that miRNA levels reported by the two platforms correlate poorly, even after correcting for potential issues inherent to signal detection algorithms. Further investigation is warranted.
