Anisotropic fluids, such as nematic liquid crystals, can form non-spherical equilibrium shapes known as tactoids. Predicting the shape of these structures as a function of material parameters is challenging and paradigmatic of a broader class of problems that combine shape and order. Here, we consider a discrete shape optimization approach with finite elements to find the configuration of two-dimensional and three-dimensional tactoids using the Landau de Gennes framework and a Q-tensor representation. Efficient solution of the resulting constrained energy minimization problem is achieved using a quasi-Newton and nested iteration algorithm. Numerical validation is performed with benchmark solutions and compared against experimental data and earlier work. We explore physically motivated subproblems, whereby the shape and order are separately held fixed, respectively, to explore the role of both and examine material parameter dependence of the convergence. Nested iteration significantly improves both the computational cost and convergence of numerical solutions of these highly deformable materials.

This work focuses on low bitrate video streaming scenarios (e.g. 50 - 200Kbps) where the video quality is severely compromised. We present a family of novel deep generative models for enhancing perceptual video quality of such streams by performing super-resolution while also removing compression artifacts. Our model, which we call SuperTran, consumes as input a single high-quality, high-resolution reference images in addition to the low-quality, low-resolution video stream. The model thus learns how to borrow or copy visual elements like textures from the reference image and fill in the remaining details from the low resolution stream in order to produce perceptually enhanced output video. The reference frame can be sent once at the start of the video session or be retrieved from a gallery. Importantly, the resulting output has substantially better detail than what has been otherwise possible with methods that only use a low resolution input such as the SuperVEGAN method. SuperTran works in real-time (up to 30 frames/sec) on the cloud alongside standard pipelines.

The protein-protein interactions (PPIs) of 14-3-3 proteins are a model system for studying PPI stabilization. The complex natural product Fusicoccin A stabilizes many 14-3-3 PPIs but is not amenable for use in SAR studies, motivating the search for more drug-like chemical matter. However, drug-like 14-3-3 PPI stabilizers enabling such study have remained elusive. An X-ray crystal structure of a PPI in complex with an extremely low potency stabilizer uncovered an unexpected non-protein interacting, ligand-chelated Mg 2+ leading to the discovery of metal ion-dependent 14-3-3 PPI stabilization potency. This originates from a novel chelation-controlled bioactive conformation stabilization effect. Metal chelation has been associated with pan-assay interference compounds (PAINS) and frequent hitter behavior, but chelation can evidently also lead to true potency gains and find use as a medicinal chemistry strategy to guide compound optimization. To demonstrate this, we exploited the effect to design the first potent, selective and drug-like 14-3-3 PPI stabilizers.

In the work, a modified effective medium theory is constructed for calculating the effective properties of thermoelectric composites with different values of percolation thresholds. It is shown that even at concentrations beyond the critical region, the threshold value is essential for determining the effective properties. Two fundamentally different cases of a set of local properties of the composite are considered. In one of these cases, the conductivity and thermal conductivity of one of the phases is simultaneously greater than the conductivity and thermal conductivity of the other phase. The second, anomalous case, when the electrical conductivity of the first phase is greater than that of the second, but the thermal conductivity of the first phase is less than that of the second, shows unusual concentration behavior of effective conductivity, i.e. with an increase in the well-conducting phase, the effective conductivity shows a decrease (rather than growth as in the standard case), which at goes over to growth.

It is discussed that the classical effective medium theory for the elastic properties of random heterogeneous materials is not congruous with the effective medium theory for the electrical conductivity. In particular, when describing the elastic and electro-conductive properties of a strongly inhomogeneous two-phase composite material, the steep rise of effective parameters occurs at different concentrations. To achieve the logical concordance between the cross-property relations, a modification of the effective medium theory of the elastic properties is introduced. It is shown that the qualitative conclusions of the theory do not change, while a possibility of describing a broader class of composite materials with various percolation thresholds arises. It is determined under what conditions there is an elastic analogue of the Dykhne formula (for the effective conductivity). The introduction of the theory with the variable percolation threshold paves the way for describing the magneto-elastic properties of magnetorheological elastomers, as it has been achieved for magneto-dielectric and magnetic properties recently.

The Black-Litterman model combines investors' personal views with historical data and gives optimal portfolio weights. In this paper we will introduce the original Black-Litterman model (section 1), we will modify the model such that it fits in a Bayesian framework by considering the investors' personal views to be a direct prior on the means of the returns and by adding a typical Inverse Wishart prior on the covariance matrix of the returns (section 2). Lastly, we will use Leonard and Hsu's (1992) idea of adding a prior on the logarithm of the covariance matrix (section 3). Sensitivity simulations for the level of confidence that the investor has in their own personal views were performed and performance of the models was assessed on a test data set consisting of returns over the month of January 2018.
